چکیده

We demonstrate a novel assay for purification of RNA (16S rRNA) from whole human blood infected with Pseudomonas Putida (P. Putida). This assay is unique in that the extraction chemistry provides a method which both lyses bacteria cells and protects RNA from degradation when isolating from matrices rich with ribonuclease (RNase) (such as blood). The method can also be integrated with on-chip RNA purification using isotachophoresis (ITP). We demonstrate that the extracted 16S rRNA is compatible with reverse transcription polymerase chain reaction (RT-qPCR) and detect P. putida infected whole blood at clinically relevant bacterial cell concentration of 30-30,000 bacteria per microliter of blood.

Microfluidic extraction of tannic acid from Quercus leaves

In this study, extraction of tannic acid using microchannel was investigated. Affective parameters were optimized. Different solvents including buthanol, ethylacetate and n-hexane as organic phase, methanol, propanol, ethanol and water as aqueous phase investigated. Microchannels with different confluence angles and diameters were examined. Microchannels with different confluence angles and dia...

Propionic acid extraction in a microfluidic system: simultaneous effects of channel diameter and fluid flow rate on the flow regime and mass transfer

In this work, extraction of propionic acid from the aqueous phase to the organic phase (1-octanol) was performed in T-junction microchannels and effects of channel diameter and fluid flow rate on the mass transfer characteristics were investigated. The two-phase flow patterns in studied microchannels with 0.4 and 0.8 mm diameters were observed. Weber ‎ number and surface-to-volume ratio were ca...

A World-to-digital Microfluidic Interface for Total Rna Extraction from Blood Samples

While clinical transcriptomics methods vary widely, a common characteristic is the need for chemical processing of extremely heterogeneous fluids (e.g., whole blood) prior to analysis. Here, we introduce the first world-to-digital microfluidic (DMF)-based method for chemical processing of RNA in blood. This sample-to-answer system has great potential for rapid characterization of novel and emer...

The challenge of getting a high quality of RNA from oocyte for gene expression study

The extraction of intact RNA from oocyte is quite challenging and time-consuming. A standard protocol using commercial RNA extraction kit, yields a low quantity of RNA in oocytes. In the past, several attempts in getting RNA for gene expression study ended up with a few different modified methods. Extraction of high-quality RNA from oocyte is important before further downstream analyses such as...

Optimization of RNA Extraction from Rat Pancreatic Tissue

Background: Optimized RNA extraction from tissues and cell lines consists of four main stages regardless of the method of extraction: 1) homogenizing, 2) effective denaturation of proteins from RNA, 3) inactivation of ribonuclease, and 4) removal of any DNA, protein, and carbohydrate contamination. Isolation of undamaged intact RNA is challenging when the related tissue contains high levels of ...
